THEMES DE RECHERCHE :
Nous nous intéressons principalement aux mécanismes moléculaires de la transmission synaptique et de l’excitabilité neuronale et la recherche dans cet axe thématique se concentre autour de 3 projets :
1- Mécanismes moléculaires du contrôle de l’excitabilité neuronale par la protéine LGI1 (Leucine-rich Glioma-Inactivated 1)
2- Activité et Récepteurs des Neurotoxines Botuliques
3- Implication des sous-unités du secteur V0 de la V-ATPase dans la modulation de transmission synaptique
1- Mécanismes moléculaires du contrôle de l’excitabilité neuronale par la protéine LGI1 (Leucine-rich Glioma-Inactivated 1)
La protéine LGI1 (leucine-rich, glioma-inactivated 1) est une glycoprotéine soluble impliquée dans deux maladies neurologiques : l’épilepsie temporale latérale autosomique dominante et certaines formes auto-immunes d’encéphalite limbique. Dans le cas de l’encéphalite limbique, l’autoimmunité se solde par la présence chez certains patients d’auto-anticorps dirigés contre la LGI1 et l’épilepsie est associée à des perturbations psychiatriques, ainsi qu’à des pertes de mémoire.
La LGI1 interagit avec les metalloprotéases inactives ADAM22 et 23. Elle est présente dans des complexes synaptiques multi-moléculaires incluant les canaux Kv1 et les récepteurs AMPA. Depuis plusieurs années, notre équipe pratique des tests d’aide au diagnostic de l’encéphalite limbique autoimmune LGI1-dépendante en utilisant l’immunoprécipitation des canaux Kv1 radiomarqués à la dendrotoxine iodée.
Des mutations dans le gène codant pour la LGI1 provoquent principalement une haplo-insuffisance due à un défaut de sécrétion de la protéine. Des crises d’épilepsies accompagnées d’aura auditifs caractérisent ainsi la majorité des patients. Les mécanismes moléculaires par lesquels l'haplo-insuffisance de LGI1 induit une perturbation de l'excitabilité neuronale sont mal connus. Nos données (Seagar et al. 2017) montrent que l’augmentation de l’excitabilité neuronale en absence de LGI1 est consécutive à une diminution de l’expression des canaux Kv1 axonaux et présynaptiques qui jouent un rôle important dans la régulation des propriétés du neurone, telles que son excitabilité, la conduction axonale et la force synaptique. 2- Activité et Récepteurs des Neurotoxines BotuliquesLes neurotoxines botuliques sécrétées par des bactéries sont à l’origine du botulisme humain et animal qui se traduit par une dysautonomie périphérique et une paralysie flasque marquée qui peut être létale. Par ailleurs, ces neurotoxines présentent un intérêt thérapeutique majeur pour le traitement d’affections neurologiques multiples. A l’aide d’outils biologiques propres au laboratoire et d’une méthode basée sur la résonance plasmonique de surface, nous avons développé au cours des dix dernières années, des dosages ultra-performants permettant de quantifier spécifiquement l’activité protéasique de plusieurs sérotypes de neurotoxines botuliques dans des sérums de patients. En collaboration avec la société IPSEN, ces tests nous ont permis de valider l’activité de lots pharmaceutiques. Par ailleurs, nous avons développé avec la société CILOA un système exosomal permettant d’exprimer dans une conformation native à la surface d’exosomes recombinants des récepteurs membranaires possédant un ou plusieurs domaines transmembranaires (Desplantes et al. 2017). Nous avons validé l’utilité de cet apport méthodologique dans l’étude pharmacologique d’un récepteur aux protéines-G et des récepteurs aux neurotoxines botuliques. Notre approche actuelle vise à déchiffrer, au travers d’approches de biologie cellulaire, de biochimie et de modélisation, les mécanismes moléculaires qui régissent la reconnaissance des neurotoxines botuliques pour leurs récepteurs neuronaux. Dans ce sens, nous avons récemment mis en évidence une interaction directe entre le domaine extracellulaire de la synaptotagmine et les gangliosides (Flores et al. 2019). Nous proposons que ce complexe représente à la surface neuronale le récepteur de la neurotoxine botulique de type B. 3- Implication des sous-unités du secteur V0 de la V-ATPase dans la modulation de transmission synaptique
La V-ATPase (ATPase à proton vacuolaire) est un nano-moteur mutli-moléculaire présent dans toutes les cellules eucaryotes. Elle est composée de l’association réversible de deux secteurs multi-moléculaires : un secteur extra-membranaire V1 qui hydrolyse l’ATP et un secteur transmembranaire V0 qui transporte les protons. La fonction primaire de ce nano-moteur est le transport actif (ATP-dépendant) de proton. Elle acidifie les compartiments intracellulaires et joue un rôle important dans de nombreux processus physiologiques. Le couplage entre l’hydrolyse de l’ATP et le transport de proton par la V-ATPase joue un rôle crucial dans l’acidification des vésicules synaptiques et ainsi dans leur charge en neurotransmetteurs. Indépendamment de son rôle dans le transport actif de proton, il s’est dégagé ces dernières années un rôle du secteur V0 de la V-ATPase dans la libération SNARE-dépendante de neurotransmetteurs. Des composants du secteur V0 de la V-ATPase interagissent avec les protéines SNAREs et interfèrent avec l’exocytose. Nous nous intéressons particulièrement à l’implication des sous unités « V0c » et « V0d » dans la libération SNARE-dépendante DiGivanni et al. 2010; Rama et al. 2019.
